生化进展课件第1页,本讲稿共19页Abstract1、Habronema muscae(Spirurida:Habronematidae)occurs in the stomach of equids,is transmitted by adult muscid dipterans and causes gastric habronemiasis2、Scanning electron microscopy(SEM)was used to study the morphological aspects of adult worms of this nematode in detail.第2页,本讲稿共19页INTRODUCTIONBackground:1.The adult worms live superficially, under the mucous layer, with only their heads buried in the mucosal spaces in the stomach.2.The main effect of Habronema in the stomach is to stimulate the secretion of large amounts of thick, stringy, tenacious mucus, with associated histological change consisting of hyper-plasia and hypertrophy of the mucus-secreting cells.Purpose:Although there have been studies evaluating the morphology of avian habronematid nematodes based on light and scanning electron microscopy, the present(SEM)study is the first to describe,in detail,H.muscae removed from a naturally infected horse.第3页,本讲稿共19页MATERIALS AND METHODSMaterials:A total of 36 worms(22 females and 14 males)were removed from the stomach of a 13-year-old horse.第4页,本讲稿共19页Methods:1.The worms were washed in 2%sodium cacodylate buffer(pH 7.2),and then cleared with lactophenol solution.2.Identification of the species was confirmed on the basis of light microscopic examination.Among examined worms,12 were identified as H.muscae(seven females and five males).3.The nematodes were put into fresh 4%glutarldehyde for 2 h,post fixed in 1%osmium tetraoxide in cacodylate buffer(pH 7.2)at 4C for 1 h and dehydrated through a series of increasing ethanol concentrations.第5页,本讲稿共19页4.They were then dried with liquid CO2 in a critical point drier apparatus.5.Each worm was placed on a numbered aluminium stab, which was coated with gold in an ioncoating sputter and viewed with a JEOL JSM-840 Scanning Electron Microscope operated at 15 kV.Photographs were taken with a digital acquisition system for analog SEMs.第6页,本讲稿共19页RESULTSFig.1 H.muscae(Spirurida,Habronematidae),removed from naturally infected horse by SEM.Another SEM study, which was carried out on Viguiera dicrurusi(Spirurida:Habronematidae)harboured by black
