2、ofallpersonnelwhomanageandconducttheperformanceevaluationofIVDMDs,particularlyforpersonnelwhoneedtheorganisationalfreedomandauthoritytoa)assessthevalidityoftestresultsanddataalreadyavailable;b)specifyperformanceclaimswhichshallbefurtherexaminedorconfirmed;c)specifyanddocumenttheevaluationplanandthet

3、estprocedures;d)preparetheevaluationreport.Themanufacturershallappointaco-ordinatorwithoverallresponsibilityoftheperformanceevaluationstudy.Theco-ordinatorshallhimselfassurethatadequateresourcesareavailable.Theinvestigatorshallensurethattheevaluationplanisfollowedathislocationandthatthestudyisapprop

4、riatelyreviewedfromanethicalpointofview.3.2DocumentationThedocumentationoftheperformanceevaluationstudyshallcontainthefilesrelatingtoclauses3to7ofthisstandardandshallbepartofthetechnicaldocumentationoftheIVDMD.3.3FinalassessmentandreviewTheco-ordinatorshallassessanddocumentwhichperformanceclaimsarem

5、et,statewhetherclaimsarenotmetandgiverecommendationsforcorrectiveactions,wherenecessary.Theresponsiblemanagementofthemanufacturershallmakesurethattheresultsoftheperformanceevaluationstudyandtherecommendationsforcorrectiveactionsarecarefullyconsideredandproperlydocumentedbeforeissuingadeclarationofco

6、nformity.4Organisationofaperformanceevaluationstudy4.1PreconditionsBeforestartingaperformanceevaluationstudyitshallbeensuredbytheco-ordinatorthata)theperformanceclaimsoftheIVDMDwhicharethesubjectofthestudyarespecified;b)theIVDMDhasbeenmanufacturedundercontrolledproductionprocessesandconditions;c)the

7、IVDMDtobeevaluatedmeetsthequalitycontrolreleasespecifications;d)asufficientnumberofsamplesoftheIVDMDcanbeprovidedduringtheentireperiodoftheperformanceevaluationstudy;e)alllegalrequirementsforperformanceevaluationstudiesaremet;f)theinvestigator(s)is(are)adequatelyskilledandtrainedtoconductthestudyand

8、thenecessaryresourcesareavailable.4.2EvaluationplanTheevaluationplanshallstatethepurposeonscientific,technicalormedicalgrounds,thescopeoftheevaluation,thestructureandorganizationofthestudyandthenumberofdevicesconcerned.Definingtheobjectiveofthestudy,theco-ordinatorshallhaveassessedwhichperformancecl

9、aimsarealreadyverifiedbydataorscientificliterature.Theevaluationplanshallbedesignedtominimisetherequirementsforinvasivesampling.InthecaseofIVDMDsforself-testingitshallbeensuredthattheevaluationplanisappropriateandacceptabletousersandtheinformationprovidedshallbeclearandeasilyunderstood.Theevaluation

10、planshallspecifya)thattheinvestigator(s)is(are)adequatelyskilledandtrainedtousetheIVDMD;b)thelistoflaboratoriesorotherinstitutionstakingpartintheperformanceevaluationstudy;forselftesting,thelocationandnumberoflaypersonsinvolved;c)thetime-table;d)thenecessaryminimumnumberofprobandsfromwhomspecimensar

11、ecollectedbyinvasiveproceduresinordertoadequatelyassesstheperformanceoftheIVDMD;e)instructionsforuseincludingadescriptionoftheconditionsofuse;f)theperformanceclaims(e.g.analyticalsensitivity,diagnosticsensitivity,analyticalspecificity,diagnosticspecificity,accuracy,repeatability,reproducibility)tobe

12、validated;g)theformatofperformancestudyrecords.4.3SitesandresourcesIngeneral,theperformancestudyprocedure(s)shallbecarriedoutunderconditionsreflectingtherelevantintendedconditionsofuse.Theco-ordinatorshalltaketheresponsibilityfortheproperconductoftheperformanceevaluationstudyatallsites.Allinvestigat

13、orsshallbenamed.Theco-ordinatorshallensureadequatecompetenceandskillatallsitesinvolvedandthatthenecessaryresourcesareavailable.WherelaypersonsareinvolvedinaperformanceevaluationstudyofanIVDMDforself-testing,thelocationofthestudyandthenumberofpersonsshallbegiven.Theco-ordinatorshallspecifythecriteria

14、fortheselectionofarepresentativepanel.EspeciallyforstudiesinvolvinglaypersonsitshallbeensuredthatthesepersonsdonotreceiveadditionalinformationontheuseoftheIVDMDapartfromthatwhichisprovidedwiththeIVDMDwhenitisplacedonthemarketbecausethecomprehensionofthemanufacturersinstructionsforuseisoneoftheimport

15、antaspectsofthestudy.ltshallalsobeensuredthattheuntrainedperson(s)donotreceiveanyadditionalinformationorhelp,e.g.fromatutor,otherthanthetrainingspecifiedandprovidedbythemanufacturerintheinstructionsforuse.4.4BasicdesigninformationTheco-ordinatorshallprovidetheinvestigator(s)withsufficientinformation

16、inordertounderstandthefunctionandapplicationoftheIVDMDand,wherenecessary,theinvestigatorshallmakehimselffamiliarwiththeIVDMDanditsapplication.TheinformationprovidedshallincludeastatementthatthedeviceinquestionconformswiththerequirementsoftheDirective98/79/ECapartfromthosetobeevaluated.4.5Experimenta

17、ldesignTheexperimentalprocedurestovalidateeachperformanceclaimsubjecttotheperformanceevaluationstudyshallbedocumentedintheevaluationplan.Specialconsiderationinperformanceevaluationstudiesofreagents/kitsshallbegiven,whereapplicable,tothefollowing:specificationoftype(e.g.serum,plasma,urine)andproperti

18、es(e.g.concentrationrange,ageandsexoftheprobandpopulation)ofspecimensappropriatetotheintendeduse;probandstobeenrolled;suitability,stabilityandvolumeofspecimensandspecimenexclusioncriteria;blindprocedures,wherenecessary;reagentstability;inclusionofcommoninterferingfactors,causedbyspecimenconditionort

19、hepathological/physiologicalstatusofthespecimendonorortreatment;conditionsforusewhichcanbereasonablyanticipated;specialattentionshallbepaidtotheconditionsofusebylaypersons;selectionofanappropriatereferencemeasurementprocedureandreferencematerialofhigherorder,whereavailable;determinationofthestatusof

20、specimens(forqualitativetestswithanominalorordinalscale);calibrationprocedures,includingtraceability,whereappropriate;appropriatemeansofcontrol;limitationsofthetest;criteriaforre-examinationanddataexclusion;availabilityofadditionalinformationconcerningthespecimenordonoriffollow-upofunexpectedresults

21、isrequired;appropriatemeasurestoreduceriskofinfectiontotheuser.Wherethestudyisintendedtovalidatetheperformanceclaimsofaninstrumentspecialconsiderationshallbegivenadditionallytothefollowing:maintenanceandcleaning;carry-overeffects;softwarevalidation.NOTEFortheinvestigationofthetechnicalaspectsofinstr

22、uments,otherstandardscanberelevant.4.6PerformancestudyrecordsTheperformancestudyrecordsshallrefertotheexperimentalproceduresintheevaluationplan;beunequivocallyidentifiable;containorrefertoallresultsandrelatedrelevantdata;bepartofthetechnicaldocumentationoftheIVDMD.Theprotectionofallconfidentialdatas

23、hallbeensured.4.7ObservationsandunexpectedoutcomesSpecialattentionshallbepaidtoobservationsandunexpectedoutcomes,e.g.dropouts,outliers,instabilityofsampleorreagentsignaletc.,non-reproducibility,non-correlationofresultstothereferenceortothediagnosticpattern,defectsorbreakdowns,softwareerrors,orerrors

24、ignals.Anydeviationfromthedefinedproceduresshallberecorded.InthecaseofIVDMDsforself-testing,theinvestigatorortutorshalldulynoteanydifficultyorquestionausermayhaveandanydeviationfromthemodeofapplicationoftheIVDMDasdescribedbythemanufacturer.Anysuchobservationshallbeproperlyrecorded.Theco-ordinatorsha

25、ll,togetherwiththeinvestigator,tracethecausewheneverpossible.Theresultshallberecordedandshallbepartoftheevaluationreport.Wherethevalidityoftheexaminationsalreadyperformedmaybequestionablebecauseofanidentifiedsourceoferrorthetestsshallberepeatedafterexclusionofthatcause.Whereamisuseormisinterpretatio

26、noftheinstructionsforusehasbeenthecauseandwhereanunexpectedriskinherenttotheproductdesignorthemodeofapplicationhasbeenidentifiedthisshallbeclearlystated.Theproposalsoftheinvestigator(s)andtheco-ordinatorforanyimprovementoftheIVDMDand/oritsapplicationshallberecorded.4.8EvaluationreportTheco-ordinator

27、shallestablishanevaluationreport.Itshallcontainadescriptionofthestudy,ananalysisoftheresultstogetherwithaconclusionontheperformanceclaimsinvestigated.Thereportshallalsodiscussanyunexpectedoutcomeswhichhaveoccurred.Itshallidentifythecausewheneverpossibleandgiverecommendationsforcorrectiveactionstobet

28、aken,wherenecessary.IfseveralstudieshavebeenconductedforoneIVDMD,asinglesummarizingreportmaybeestablished.5ModificationsduringtheperformanceevaluationstudyWherethemanufacturingprocesshasbeenchangeditshallbecheckedwhethertheperformanceclaimsoftheIVDMDstillconformtothosewhichhadbeensetinitially.Otherw

29、isethevalidityoftheexaminationsalreadyperformedshallbequestionedandtheevaluationplanshallberevisedaccordingly.Wheredesignchangesareintroduced,theevaluationplanshallberevised.6Re-evaluationIncaseofchangestothedesignormanufacturingprocessoftheIVDMD,theperformanceevaluationstudyshallberepeatedasfarasne

30、cessary,toensurethattheintendeduseandtheperformanceclaimsoftheIVDMDplacedonthemarketareadequatelyevaluated.Thisre-evaluationmayrefertodocumentedresultsofaprecedingevaluationinsofarastheseareconsideredvalidandtransferableaftercriticalreview.7ProtectionandsafetyofprobandsTheremoval,collectionanduseoft

31、issues,cellsandsubstancesofhumanoriginisgoverned,inrelationtoethics,bytheprincipleslaiddownintheConventionoftheCouncilofEuropefortheprotectionofhumanrightsanddignityofthehumanbeingwithregardtotheapplicationofbiologyandmedicineandbyanynationalregulationsonthismatter.Inanycase,theresultsobtainedfromas

32、pecimenbymeansoftheIVDMDunderevaluationshallnotbeusedforotherpurposesthanforperformanceevaluation,unlessethicalreasons,fullysupportedbyaresponsiblemedicalprofessional,suggestthecontrary.Insuchacasethemedicalprofessionalassumescompleteresponsibility.3性能评估的一般要求3.1责任和谋略生产商负责性能评估研究的开始和/或引导。他应规范明确所有管理和引导

39、面的研究者，还有其他的可能相关的标准。4.6性能研究记录记录应：-参照评估计划中的实验过程-毫不含糊地确认-包括或参照所有结果及相关数据。-成为IVDMD的技术文档的一部分。机密数据的保护需要确保。4.7观测和非期待的结果需要特别的注意观测与不符的结果，例如，丢弃的结果，外层的结果，样品或试剂的不稳定性信号等等，不可重复性，结果对参考值或对诊断模式的不相关性，缺陷或故障，软件错误，或误差信号。任何对确切的（实验）过程的偏离都应被记录。在自测IVDMD的案例中，研究者或导师应准确标注使用者可能遇到的问题及困难，以及对IVDMD生产商的使用指南所描述的使用方式的偏差。所有的这些观测应被正确记录。可
