1、共生食用菌松茸菌丝体液体发酵的研究_菌丝体是食用菌的什么 摘要:目的探讨松茸菌丝体液体发酵方法和最佳条件。方法经RAPD-PCR 分析确认松茸菌丝体菌种,探讨松茸液体深层发酵过程中菌丝体的生物量、残糖浓度和pH随时间改变的过程;比较不同种龄对菌丝体积累的影响,确立最佳接种种龄;比较不同初始糖浓度发酵过程的结果,运用中间补糖的方法,确定最优的补糖时间。结果最佳接种种龄为6 d;初始糖浓度较低有利于缩短松茸种子液生长的延迟期,较高则可使菌丝体在较长时间保持生长并获得较高的生物量产出。d 12 为最优补糖时间,在摇瓶体系中培育24 d最终得到12.77 g/L的菌丝体生物量。结论最佳培育条件下,采纳

4、nvestigated with Tricholoma matsutake stain, which was isolated and identified by RAPD-PCR method. By comparing the effect of different inoculum ages on biomass, the optimum inoculum age was determined. Based on comparing the effect of different initial glucose concentrations on dynamic changes of f
5、ermentation, the optimum time of feeding glucose was established by carrying out the feeding culture strategy in submerged culture. ResultsThe optimum inoculum age was 6 days. The lower initial glucose concentration obviously availed to shorten the lag phase of the growing of mycelium. And the highe
6、r initial glucose concentration made the mycelium maintain growth for a long period of time and finally reached a higher mycelium yield. The d 12 was confirmed as the optimum feeding time. By applying the feeding culture strategy, 12.77g/L mycelium biomass was obtained on day 24 in flask system. Con
7、clusion The method and optimum conditions is feasible for accumulating the mycelium of Tricholoma matsutake in submerged culture. Key words:Tricholoma matsutake; mycelium; inoculum age; feeding culture; submerged culture 松茸(Tricholoma matsutake)是与松树共生的珍稀食、药用外生菌根真菌,有很高的养分价值和特别的药用效果。由于野生松茸资源已经日渐枯竭,人工培
8、育尚无法获得松茸的子实体,因此液体深层培育松茸菌丝体,提取生理活性物质并进行深加工越来越受到关注。目前对松茸菌丝体培育的探讨主要集中在松茸菌丝的养分生理、液体条件下培育基成分的优化以及培育条件的考察等方面1-3。本试验采纳了经RAPD-PCR 分析确认的松茸菌丝体菌种,探讨了松茸在摇瓶中液体条件下各生长参数的动态改变。发酵过程中采纳中间补料的方法,获得了较高的菌丝体生物量,为大规模液体深层发酵生产供应了依据。 1材料与方法 1.1菌种来源 采集我国东北延边地区产的松茸子实体,经组织分别获得菌丝体。纯培育后,经RAPD-PCR(随机引物聚合酶链反应) 分析DNA进行亲菌鉴定,确认为松口蘑菌丝体菌
9、种4。 1.2培育基 PDA 培育基用于斜面菌种培育。液体种子培育基(g/L)是葡萄糖20 , 酵母浸粉1,蛋白胨2,NH4Cl 1,KH2PO4 0.5,MgSO47H2O 0.5,维生素B1 0.05,pH自然;发酵培育基(g/L)是葡萄糖(据试验要求而定),酵母浸粉2,蛋白胨2,KH2PO4 0.5,MgSO47H2O 0.5,维生素B1 0.05,pH 5.0。 1.3方法 1.3.1培育方法液体种子培育:用直径10 mm的打孔器将生长旺盛的固体琼脂菌苔切成大小一样的菌块,接种灭菌后的液体种子培育基,菌块要薄以使其漂移在液面上。用500 mL 三角瓶,装液量100 mL ,22 ,50
