7、pheral bloodmono2nuclear cells(PBMC)of rheumatoid arthritis(RA)patients and to evaluate the relevant clinical significance.M ethodsConventionaland nested SYBR green I fluorescent dye realtime PCR was used to detect the expression of TGF R II,Smad4,and Smad7 mRNA.ResultsCompared with those of 29 heal
8、thy persons,the expressions of TGF R II and Smad4 mRNA,and the ratio of Smad4/Smad7in PBMC of 58 RA patientswere significantly increased(P0.01).ThemRNA of TGF R IIand Smad4,and Smad4/Smad7 in activeRA patients and before treatment were respectively higher than those in inactive patients and after tr
9、eatment RA.They were also in2creased markedly in III disease stage(P 0.01).There were positive correlations between Smad4 and TGF R II and the ratio ofSmad4/Smad7(r1=0.216,P0.05;r2=0.279,P0.01).ConclusionsTGF/Smad signaling pathways are enhanced in RA.De2tection of expression of the genes associated
