10、肽键水解；（2）含有一个在酶催化中起关键作用的丝氨酸；（3）活性部位都有Asp，His和Ser，它们总是簇聚在一起；（4）在活性部位Ser附近总是有一个“口袋”；（5）催化机制相同。不同点：专一性不同，即作用肽键的位点不同。这种不同是由于“口袋”的形状和电荷不同引起。例如胰凝乳蛋白酶（chymotrypsin）的口袋比较宽和深，侧面有疏水残基，以便容纳苯丙氨酸一类疏水的芳香侧链。第16页，本讲稿共19页2.用NMR揭示某酶的His12的pKa=4.0。问（1）His12处在什么环境中？（2）为什么该环境能改变His12的pKa？回答：（1）该His 12位于疏水的或非极性环境中。（2）His+

11、His+H+，pKa=-logHis+/HisH+，疏水环境改变了酸式解离平衡，有利于His12的咪唑基团不带电荷，即His增加。因此其pKa降低。第17页，本讲稿共19页3。鸡蛋清溶菌酶有两个催化基团，即Asp52和Glu35，下图是该溶菌酶的Vmax/VmaxpH作图。根据该图回答下列问题：（1）指出图中以1、2、3标出的各点的Asp52和Glu35的正确的解离状态；（2）为什么在第2点时溶菌酶的表观速度Vmax达到最大？（3）根据该图可以获知溶菌酶什么信息？为什么认为Glu35是质子化的？第18页，本讲稿共19页回答：（1）第1点：Asp-COOH，Glu-COOH，；第2点：Asp-C

12、OO-，Glu-COOH；第3点：Asp-COO-，Glu-COO-；（2）根据phillips等人的研究，溶菌酶催化胞壁多糖水解时结合多糖的六糖单位，并特异地在D-E间水解糖苷键，该过程需要Glu35提供质子给D-E间糖苷键的O原子，促使D-E间在D环的C1O处断裂，形成正碳离子过渡态中间产物。带负电荷的Asp52能稳定带正电荷的碳阳离子过渡态。此外Asp52在一个复杂的氢键网络中也可能起氢键受体作用，根据（1）分析，pH5.0时正好Glu质子化（Glu-COOH），可以作为质子供体，Asp去质子化（Asp-COO-），故该pH使溶菌酶有最大表观速度。（3）根据Asp52和Glu35是催化基团，pH显著影响酶的活性，说明质子传递和氢键网络（广义酸碱催化）起着重要的作用。根据对溶菌酶水解底物D-E键周围的微环境的分析，知道Asp52和Glu35分别位于该糖苷键的两侧，因此两个酸性侧链具有明显不同的微环境。Glu35和D-E间O原子的距离为0.3nm，正是合适的作用距离。因此可以推测在溶菌酶最适pH（5.0）条件下，Glu35在非极性环境，质子化（Glu-COOH），Asp52处在极性环境，去质子化（Asp-COO-）。第19页，本讲稿共19页
